Titre : Interactions between solubilized membrane transporters

Only a limited number of high resolution 3D structures of membrane proteins
are available. Among other reasons, crystals of membrane proteins are hard
to obtain. For soluble proteins, favourable crystallisation conditions are
found when precipitant leads to weak inter-macromolecular attractions. For
membrane proteins, general rules may consider the presence of detergent in
the solution. We propose to study the relations between the concentration of
species in solutions (protein, free detergent, precipitant, lipids....),
their interactions (association state, solvation weak interparticle
interactions) and capacity of the membrane proteins to crystallize. Neutron
scattering and analytical ultracentrifugation combined with specific
deuterium labelling will be used in order to distinguish detergent and
protein components. Different membrane transporters involved on the
transport of drugs will be studied.

 

Among other possibilities, the subject is proposed by the International PhD
program of the Life Sciences (DSV) of the Commissariat à l’Energie Atomique
(CEA) (deadline for on line application: Mars 8th 2008:

2008. Duration: three years. The gross annual salary is between 18300 and
23000 Euros.
