Hi Dror,
The most obvious consideration is the removal or RNases from the cells
and other plasticware, etc that will come into contact with the sample.
We had success doing an equilibrium experiment whereby we soaked the
windows and centerpieces in 1 M HCl for 2 hours, rinsed with DEPC
treated water, soaked again with 1 M NaOH for 2 hours, rinsed again with
DEPC treated water and rinsed with methanol before assembling the cell.
The pipet tips and tubes were autoclaved for 45 minutes. Another method
is to treat the individual pieces with a commercial RNase inhibitor such
as RNaseZap available from Ambion according to their directions.
Good luck and Happy Holidays to all,
Virgil
