Dear collleauges, dear Roy,
To run 6 sector cells in the AUC is for several reasons a bit a delicate /tricky run. Previius models tend to leak between the sectors especially at higher speeds, thatfore Beckman limited the rotor speed., (but even then 20% off was better). Beckman recomaned to add the FC-43
fluid (Flouro - carbo)to obtain a bottom-phase that produce a marked "bottom-meniscus, clear seen in any optic system used.It is a good auxillary methode for radius determination, and helps also to test , if their was no leak from an upper or lower sector. At least it is an instrument to test leaking cells, not only the 6-channel one.
Sorry A) I couldn't find the reference of the right Beckman bulletin publication B)sure that FC43 can be used only with aqueaous solutions that do not influence the protein.
Yours....ariel
