[RASMB] UltraScan AccessGrid Seminar on Thursday

Dear Colleagues,

I would like to announce the next UltraScan AccessGrid seminar which
will take place on Thursday, December 3rd, from 11 am - 12:30 pm
Central Time in the UTHSCSA UltraScan AccessGrid venue (England time:
5 pm, Central Europe: 6 pm).

This seminar will be presented by Dr. Susumu Uchiyama, Osaka University,
Japan, who will report on antibody-antigen interactions studied by
analytical ultracentrifugation and mass spectrometry.

Abstract:

We have recently employed the combination of two different biophysical
methods, analytical ultracentrifugation (AUC) and mass spectrometry under
non-denaturing condition (MS), for the identification of components in
heterogeneous protein solution. MS provides the information on each
component with high molecular weight resolution, whereas AUC gives
information on components in solution. In this seminar, the study on the
interactions between monoclonal antibody (Ab) and its antigen (Ag) will
be introduced. Solutions of two anti-hapten Abs with different affinities
mixed with hapten-conjugated BSA were analyzed. The results obtained
from AUC sedimentation velocity (AUC-SV) and MS were consistent with each
other, showing the presence of the large complex only in the solution of
high affinity Ab and Ag. ULTRASCAN GA-MC analysis of AUC-SV results for
the mixed solution of high affinity Ab and Ag clarified stoichiometry
and molecular shape of each component. Our current study indicates
that AUC and MS are two complementary methods providing unambiguous and
quantitative information on the interactions among molecules in solution.

The UltraScan seminar is broadcast over the AccessGrid. In order to
participate you will need to have AccessGrid 3.0.2 or higher installed
on your computer. If you are not on a multicast network (most likely
you will not have this capability), you need to connect via unicast,
and you will need to use a unicast/multicast bridge to communicate
(Tools:Bridges in the Venueclient menu). You can use the following
MC-bridges to connect to our venueserver: Argonne, NCSA and the UTHSCSA
bridge. Participants are strongly encouraged to test their connections
one or two days in advance with Jeremy Mann (jeremy at biochem.uthscsa.edu)
